Adult Monotherapy Up to 200 mg/m2 as continuous IV infusion for 5 days repeated at approx 2-wkly intervals. Meningeal leukaemia 30 mg/m2 intrathecally every 4 days until CSF findings are normal, followed by 1 additional treatment. Dose range: 5-75 mg/m2 once daily for 4 days to once every 4 days.
Anaphylactic reactions. Bone marrow suppression w/ leukopenia, thrombocytopenia & anaemia; preexisting drug-induced bone marrow suppression. Intrathecal use may increased risk of spinal cord toxicity. Acute pancreatitis; vomiting; conventional & experimental dose schedules. Periodic checking of bone marrow exam, liver & kidney functions. Monitor blood uric acid levels. Avoid use w/ live vaccines. Preexisting hepatic dysfunction or poor renal function. Not recommended during pregnancy. Lactation. Severe neurological AR in juveniles & adolescents.
